Principal Scientist -Discovery Attribute Sciences
Posted on Apr 6, 2021 by Amgen
Let's do this. Let's change the world. In this vital role you will be a aprt of the Discovery Attribute Sciences group providing scientific and technical expertise to support early discovery research through to later stage molecule optimization and late-stage selection of Amgen's pipeline of drug candidates. This position is an exciting opportunity to unlock the potential of biology for patients suffering from serious illnesses by discovering, developing, manufacturing and delivering innovative human therapeutics.
Identifying and characterizing attributes of therapeutic protein candidates utilizing multiple analytical techniques.
Develop and apply novel sample preparation and analytical methods for analysis of therapeutic proteins and synthetic molecules with emphasis on mass spectrometry (LC-MS, LC-MS/MS and MALDI-MS), capillary electrophoresis (and CE-MS), cation exchange (and CEX-MS), size exclusion chromatography (and SEC-MS) and other biophysical techniques such as calorimetry and rheology.
Experience with LC-MS/MS to map and characterize post translational modifications such as deamidation, isomerization and oxidation would be advantageous.
Investigation, development and the application of miniaturized and high throughput mass spectrometric methods is expected.
This role will also involve supervising and mentoring a small group of MS-focused scientists. Importantly, the expectation for this role is also to be 75% laboratory based, providing support for multiple projects and their advancement and also be able to perform innovative MS application and method development.
Experience with small molecule MS and MS/MS accurate mass measurements, empirical formulae derivation and structural characterizations would be advantageous.
Communicate (verbal and in written) scientific concepts and results.
Work collaboratively with other scientists on multidisciplinary teams.
Work independently in advancing projects and studies within required deadlines.
Actively create or contribute to patent and publications.
Keep current with field of expertise, participate and advance drug development, technologies and information services programs within Amgen.
Demonstrate external scientific eminence in the form of peer-reviewed publications and conference presentations.
Follow protocols and procedures, maintain accurate records in an electronic lab notebook, and generate quality data for use in id
We are all different, yet we all use our unique contributions to serve patients. The Scientific professional we seek is a proven leader with these qualifications.
A doctorate degree with 7-10+ years in Analytical Chemistry, Biochemistry or equivalent with emphasis in mass spectrometry and other analytical characterization techniques, such as reversed-phase, size exclusion chromatography, capillary electrophoresis.
Relevant experience working as part of a multidisciplinary team in a biopharmaceutical setting would also be an advantage.
Documented theoretical knowledge and hands-on experience in peptide mapping of proteins, intact and reduced LC-MS analysis for identification and quantitation of chemical modifications, native mass spectrometry, ion mobility and experience with different fragmentation techniques such as ETD, ECD and HCD.
Training and experience of operating Thermo Orbitrap, Agilent and Waters Q-ToF mass spectrometers, Waters and Agilent UPLCs would be advantageous. Proficiency with Microsoft Office.
Knowledge of statistical processing of data, including related software like Excel, Spotfire, JMP or ArrayStudio and multiple mass spectrometric (MassLynx, MassHunter, XCalibur) and chromatographic software (ChemStation) control and data processing (MaxEnt, PMI Intact) packages is required.
